The Tunisian Ligue Professionnelle 1 edged out Al Hilal 3-2 on aggregate having won 3-1 at home in the first-leg clash.
Experienced forward Oussama Darragi will be keen to inspire Africain to victory against Constantine.
Meanwhile, Constantine eased into the group stage after thrashing Ugandan outfit Vipers SC in the first round late last month.
The Algerian Ligue Professionnelle 1 champions beat Vipers 3-0 on aggregate - winning both home and away.
Mohamed Walid Bencherifa, who netted against Vipers, is the dangerman for Constantine.
Gabonese referee Eric Otogo-Castane will officiate the game between Africain and Constantine.
